Understanding the Mouse Brain for AI Advancement
The study of the fly connectome has greatly changed neuroscience by revealing how brain networks work. Now, applying this knowledge to the mouse brain, which is more similar to the human brain, can lead to amazing advancements. Challenges in Mapping the Mouse Brain
Mapping the larger mouse brain is a tough task. Current methods can make it very expensive—more than the Human Genome Project—due to the extensive proofreading needed. This proofreading process, which corrects AI-generated data, consumes over 95% of the project costs. To make mapping the mouse brain more practical and affordable, we need to minimize human proofreading efforts. This will allow us to explore various brain states and disorders effectively.
Introducing PRISM: A Game-Changer in Brain Connectomics
Researchers from E11 Bio, with partners like the Crick Institute and MIT, have launched PRISM (Photo-connectomic Reconstruction by Iterative Staining with Molecular annotations). This innovative platform makes brain connectomics faster, cheaper, and more scalable by assigning unique protein barcodes to cells. This allows AI to self-proofread, drastically cutting down the need for human involvement and reducing costs by over 95%. PRISM combines light-sheet microscopy and expansion techniques to deliver high-resolution images at a lower cost, leading to a potential 100-fold decrease in whole-brain mapping expenses.
How PRISM Works
E11 Bio’s PRISM platform enhances brain connectomics through:
- Genetic protein barcoding: This gives neurons unique identifiers for better cell distinction.
- Expansion microscopy: It uses light microscopy for nanoscale imaging, lowering the dependence on expensive electron microscopy.
- Self-proofreading AI: Advanced AI models utilize data for precise image segmentation, reducing the need for manual corrections.
PRISM aims to map entire mammalian brains in the next five years, continuously enhancing imaging and segmentation while collaborating globally to advance neuroscience and AI technologies.
